In Vitro Propagation of Ipomoea pes-caprae (Railroad-Vine)

Abstract

A plant micropropagation protocol for in vitro establishment, plant multiplication, rooting, and ex vitro acclimatization of the dune species Ipomoea pes-caprae (Railroad-vine) was developed. Supplementation of agar-solidified Murashige and Skoog medium (MS) with either the cytokinin 2- isopentenyladenine (2iP) or zeatin significantly promoted shoot regeneration from nodal segments. Maximum shoot regeneration occurred from nodal segments cultured on agar-solidified MS supplemented with 5 mg/liter 2iP. Medium supplementation with either 2iP or zeatin did not promote axillary branching of the shoots produced. Microcuttings were effectively rooted (95%) in vitro when cultured on agar-solidified full-strength MS supplemented with 1.0 mg/liter indolebutyric acid (lBA). Transplant survival of rooted I. pes-caprae microcuttings was 100%. Approximately 446,000 I. pes-caprae plants can be produced per year from a single nodal segment using this procedure
