A preliminary study on pothos regeneration

Abstract

In vitro adventitious shoot regeneration was achieved from pothos leaf and leaf petiole explants collected from three year-old greenhouse grown cultivars of 'Jade' and 'Golden'. Explants were cultured in MS medium supplemented with growth regulators of y-naphthaleneacetic acid (NAA) with 1-phenyl-3- (1,2,3 thiadiazol-5-yl) urea (thidiazuron, TDZ), 6-(4-hydroxy-3-methylbut-2-enylaminol) purine (zeatin) or ^-(y-ydimethylallylamino)
purine (2iP). Regeneration of 'Jade' and Golden' occurred 30 and 40 days respectively from media containing TDZ, but 50 and 60 days in media containing 2iP orzeatin. Callus and regeneration initially occurred from the cut surface of the midrib or the surface area close to the midrib. Regeneration rates from leaf and leaf petiole explants were approximately 10% and 30%, respectively.
